Mebendazole functions as a microtubule polymerization inhibitor and serves as an anthelmintic agent. In cell-free assays, it demonstrates a reduction in microtubule polymerization at a concentration of 10 µM. Additionally, it impacts the viability of Giardia duodenalis by decreasing container attachment, with a half-maximal inhibition of binding (IB50) measured at 190 nM. Mebendazole also inhibits the growth of SK-MEL-19 and M14 melanoma cell lines, showing IC50 values of 300 and 320 µM, respectively. At a concentration of 1 µM, it triggers apoptosis and poly(ADP-ribose) polymerase (PARP) cleavage in these melanoma cells, while sparing melanocytes. Furthermore, it mitigates oxidative stress-induced cell death in primary mouse cortical neurons in a dose-dependent manner. In vivo studies reveal that administering mebendazole (800 µg per animal every other day) leads to a reduction in tumor volume, weight, hemoglobin levels, and vascularization in an H460 lung cancer mouse xenograft model. Additionally, a dosage of 8.8 mg/kg effectively eliminates the worm burden of Strongylus vulgaris, Strongylus edentatus, and Strongylus equinus in infected ponies. Mebendazole-containing formulations have been utilized for treating gastrointestinal infections caused by helminths.
